Data Synthesis Improves 3D Myotube Instance Segmentation

Myotubes are multinucleated muscle fibers serving as key model systems for studying muscle physiology, disease mechanisms, and drug responses. Mechanistic studies and drug screening thereby rely on quantitative morphological readouts such as diameter, length, and branching degree, which in turn require precise three-dimensional instance segmentation. Yet established pretrained biomedical segmentation models fail to generalize to this domain due to the absence of large annotated myotube datasets. We introduce a geometry-driven synthesis pipeline that models individual myotubes via polynomial centerlines, locally varying radii, branching structures, and ellipsoidal end caps derived from real microscopy observations. Synthetic volumes are rendered with realistic noise, optical artifacts, and CycleGAN-based Domain Adaptation (DA). A compact 3D U-Net with self-supervised encoder pretraining, trained exclusively on synthetic data, achieves a mean IPQ of 0.22 on real data, significantly outperforming three established zero-shot segmentation models, demonstrating that biophysics-driven synthesis enables effective instance segmentation in annotation-scarce biomedical domains.

Low-Dose CT for Stroke Diagnosis: A Dual-Pipeline Deep Learning Framework for Portable Neuroimaging

Portable CT scanners may support earlier stroke assessment, but reduced photon counts introduce noise that affects image quality and may alter automated classification. We compared direct classification of simulated low-dose slices with residual U-Net denoising followed by the same fixed classifier. Poisson noise was generated at photon-count scaling factors of 1, 5, 10, 20, and 40 using three deterministic seeds. The held-out set contained 809 slices, including 242 positive and 567 negative slices. Direct classification reached its highest mean ROC-AUC at a scaling factor of 20 (0.937 +/- 0.002). Denoising followed by classification reached 0.842 +/- 0.005 at a scaling factor of 1 and declined to 0.655 +/- 0.002 at a scaling factor of 40. Meanwhile, reconstruction quality rose steadily from 21.92 to 40.91 dB PSNR and from 0.761 to 0.987 SSIM. Both pathways had poor sensitivity at a fixed 0.5 cutoff because their classification scores were concentrated near zero. Patient-level analysis was limited by the test-set composition: all 10 patients were positive under the mask-derived patient label, preventing estimation of patient-level AUC and specificity. Overall, higher reconstruction fidelity did not translate into better discrimination by the fixed classifier except at the lowest photon-count setting.

Rethinking Garment Conditioning in Diffusion-based Virtual Try-On: Decouple, Don't Denoise

Virtual Try-On (VTON) synthesizes realistic images of a person wearing a target garment, with broad applications in e-commerce and fashion. Diffusion-based dual-UNet methods achieve strong results but double the parameters by dedicating a separate network to garment conditioning. Spatial concatenation offers a simpler single-network alternative, yet both UNet- and DiT-based instantiations report that full fine-tuning is ineffective, and the community has settled for attention-only training. We ask: why does full fine-tuning fail, and can this be resolved? Through what is, to our knowledge, the first visualization study of dual-UNet reference network behavior, we identify a unifying insight: garment conditioning must be decoupled from the denoising process. Spatial concatenation violates this by embedding the garment within the denoising target, causing three conflicts: guidance leakage, gradient competition, and train-test discrepancy. We derive three design principles to restore this decoupling and implement them as a pure recipe atop a standard architecture with no modification. The resulting model, DeCo-VTON (860M params), achieves single-network state of the art, matching the dual-UNet state of the art at half the cost while being preferred in human evaluation.

Segmentation and Classification of Pap Smear Images for Cervical Cancer Detection Using Deep Learning

Cervical cancer remains a significant global health concern and a leading cause of cancer-related deaths among women. Early detection through Pap smear tests is essential to reduce mortality rates; however, the manual examination is time consuming and prone to human error. This study proposes a deep learning framework that integrates U-Net for segmentation and a classification model to enhance diagnostic performance. The Herlev Pap Smear Dataset, a publicly available cervical cell dataset, was utilized for training and evaluation. The impact of segmentation on classification performance was evaluated by comparing the model trained on segmented images and another trained on non-segmented images. Experimental results showed that the use of segmented images marginally improved the model performance on precision (about 0.41 percent higher) and F1-score (about 1.30 percent higher), which suggests a slightly more balanced classification performance. While segmentation helps in feature extraction, the results showed that its impact on classification performance appears to be limited. The proposed framework offers a supplemental tool for clinical applications, which may aid pathologists in early diagnosis.

Converting T1-weighted MRI from 3T to 7T quality using deep learning

Ultra-high resolution 7 tesla (7T) magnetic resonance imaging (MRI) provides detailed anatomical views, offering better signal-to-noise ratio, resolution and tissue contrast than 3T MRI, though at the cost of accessibility. We present an advanced deep learning model for synthesizing 7T brain MRI from 3T brain MRI. Paired 7T and 3T T1-weighted images were acquired from 172 participants (124 cognitively unimpaired, 48 impaired) from the Swedish BioFINDER-2 study. To synthesize 7T MRI from 3T images, we trained two models: a specialized U-Net, and a U-Net integrated with a generative adversarial network (GAN U-Net). Our models outperformed two previous state-of-the-art 3T-to-7T models in image-based evaluation metrics. Four blinded MRI professionals judged our synthetic 7T images as comparable in detail to real 7T images, and superior in subjective visual quality to 7T images, due to the reduction of artifacts. Using both SynthSeg and NextBrain, automated segmentations of the synthetic 7T images were more similar to real 7T segmentations than automated segmentations from the 3T images that were used to synthesize the 7T images. Finally, synthetic 7T images showed similar performance to real 3T images in downstream prediction of cognitive status using MRI derivatives (n=3,168). In all, we show that synthetic T1-weighted brain images approaching 7T quality can be generated from 3T images, which may improve image quality and segmentation, without compromising performance in downstream tasks. Future directions, possible clinical use cases, and limitations are discussed.

Fast Equivariant Imaging: Accelerating Unsupervised Learning and Model Adaptation via Inexact Splitting

In this work, we propose Fast Equivariant Imaging (FEI), a novel unsupervised learning framework to rapidly and efficiently train deep imaging networks without ground-truth data. FEI reformulates the EI objective through an inexact variable-splitting scheme, decoupling network training from an auxiliary restoration step implemented with a plug-and-play denoiser, this novel unsupervised scheme shows superior efficiency and performance compared to the standard Equivariant Imaging paradigm. In particular, our FEI schemes achieve an order-of-magnitude (10x) acceleration over standard EI on training U-Net for X-ray CT reconstruction and image inpainting, with improved generalization performance. Beyond offline training, the proposed scheme also enables efficient test-time adaptation of a pretrained model to individual samples, to secure further performance improvements. Extensive experiments show that the proposed approach provides a noticeable efficiency and performance gain over existing unsupervised methods and model adaptation techniques.

MSA-UNet3+: Multi-Scale Attention UNet3+ with New Supervised Prototypical Contrastive Loss for Coronary DSA Image Segmentation

Accurate segmentation of coronary Digital Subtraction Angiography (DSA) images is essential for diagnosing and treating coronary artery disease (CAD). Despite advances in deep learning, challenges such as high intra-class variance and class imbalance limit precise vessel delineation. Existing approaches for coronary DSA segmentation cannot effectively address these issues. Furthermore, existing segmentation network encoders do not directly generate semantic embeddings, which could enable the decoder to reconstruct segmentation masks more effectively. We propose a Supervised Prototypical Contrastive Loss (SPCL) that combines supervised and prototypical contrastive learning to enhance coronary DSA image segmentation. The supervised contrastive loss enforces semantic embeddings in the encoder, improving feature differentiation. The prototypical contrastive loss enables the model to focus on the foreground class while alleviating high intra-class variance and class imbalance by concentrating only on hard-to-classify background samples. We implement the proposed SPCL within MSA-UNet3+, a Multi-Scale Attention-Enhanced UNet3+ architecture. The architecture integrates a Multi-Scale Attention Encoder (M-encoder), a Multi-Scale Dilated Bottleneck (MSD-Bottleneck) for multi-scale feature extraction, and a Contextual Attention Fusion Module (CAFM) to preserve fine-grained details while improving contextual understanding. Experiments on a private coronary DSA dataset demonstrate that MSA-UNet3+ outperforms state-of-the-art methods, achieving the highest Dice coefficient and F1-score while significantly reducing ASD and ACD. The framework provides precise vessel segmentation for accurate identification of coronary stenosis and supports informed diagnostic and therapeutic decisions. UniReg: Conditional Unified Model for Medical Image Registration

Learning-based medical image registration has matched the accuracy of conventional methods while offering superior computational efficiency. However, existing approaches suffer from poor generalization across diverse clinical scenarios, requiring the laborious development of multiple isolated networks for specific registration tasks, \emph{e.g.}, inter-/intra-subject registration or anatomical region-specific alignment, leading to cumbersome development pipelines. To overcome this limitation, we propose \textbf{UniReg}, the first conditional unified model for multi-scenario medical image registration, which combines the precision advantages of task-specific learning methods with the generalization of traditional optimization methods. Our key innovation is a unified registration framework that adaptively estimates deformation fields conditioned on: (1) anatomical structure priors, (2) registration type constraints (inter/intra-subject), and (3) instance-specific features, enabling effective alignment across heterogeneous CT and MR registration scenarios within a single model. Through comprehensive experiments on multiple CT/MR registration datasets, UniReg achieves superior average registration accuracy compared with current state-of-the-art learning-based methods while exhibiting strong cross-scenario generalization. Moreover, by replacing multiple isolated task-specific models with a compact unified model, UniReg substantially reduces the overall training burden in terms of total training cost and model redundancy.
